We plan to obtain draft genome sequences of 150 LA-MRSA strains isolated from human, animal, and environmental samples collected at various US swine farms. Next, we plan to compare the draft genome sequences obtained for these LA-MRSA strains to each other and to other MSSA and MRSA publicly available genome sequences to generate phylogenetic relationships between strains, antibiotic resistant genes, and mobile genetic elements carried on phages for the purpose of gaining a more comprehensive assessment of the origin, evolution, and zoonotic potential of these strains. Finally, we plan to use in vitro binding assays to test the capacity of the LA-MRSA strains, which draft genome sequence information will be obtained, to adhere to human and porcine keratinocytes.
