Objective:
Utilize RNA-Seq technology to develop horn fly tissue-specific transcriptome databases. The sequence information obtained will allow for identification of genes differentially expressed between selected developmental stages and ultimately advance our understanding of pathways critical to horn fly olfaction and insect-microbe interactions.

Approach:
Construct ten TruSeq RNASeq libraries using biological replicates of five different horn fly tissues. The samples will undergo 2x100 base pair paired-end sequencing on a HiSeq2000 instrument, the data of which will be analyzed using bioinformatic tools to assemble de novo horn fly transcriptomes and to conduct specific gene expression profiling between selected experimental conditions.
