Objective:
1. Create six recombinant inbred line (RIL) populations in potato. 2. Phenotype and genotype the RIL populations and their founders. 3. Produce homozygous diploid cultivated potato clones.

Approach:
Objective 1: Create recombinant inbred line (RIL) populations in potato. We will create a set of six genetically diverse recombinant inbred line populations (RILs). The six female founder parents of the RILs carry cultivated and wild species genes for processing quality, tuber yield and size, tuber color and shape, tuber dormancy, and resistance to major pathogens. The common male parent for all RILs will be M6, an inbred S. chacoense line homozygous for Sli. Objective 2: Phenotype and genotype the RIL populations and their founders. Each of the 600 lines (6 populations x 100 lines per population) will be grown as families of 10 individuals per line. We will evaluate each family for plant vigor, vine uniformity, vine maturity, flowering, tuber traits (size, shape, number, skin color, flesh color, dry matter content, chip color). In addition, each family will be genotyped and the founder parents will be sequenced. Objective 3. Develop inbred lines of cultivated potato. While the S. chacoense inbred line M6 will provide the backbone for Objectives 1 and 2, a concurrent project to develop inbred cultivated potato (S. tuberosum) will be carried out in Objective 3. We will attempt to create monoploids and then double them to produce homozygous lines.
