Technical Abstract: Optimization of extraction, fractionation, and reaction conditions using critical fluids can be a time consuming and labour-intensive process. In this study, automated computer controlled extraction instrumentation has been used in a combinatorial mode to establishment of the best chromatographic conditions for fractionating phospholipids obtained from soybeans. The described approach and equipment allows the automated extractor to screen the effectiveness of individual sorbents for separating and concentrating the phospholipid moieties, while at the same time altering the properties (pressure, temperature, cosolvent, etc.) of the mobile phase to affect elution. Similarly, a number of pressures and temperatures were examined rapidly using this approach to optimize the extraction of oil from cedarwood using supercritical carbon dioxide (SC-CO2). For the latter natural product matrix, it was shown that the cedarwood oil can be very rapidly extracted and that rapid extraction is required to maintain the compositional integrity of the original oil. SC-CO2 was more effective in removing the oil relative to a traditional steam distillation method which usually only recovers about 50% of the available oil. These conclusions and optimization of the above described processes can be rapidly facilitated using the described equipment.
