Interpretive Summary:

Technical Abstract: ISTA has initiated a ring test to compare methods for the detection of Clavibacter michiganense subsp. insidiosum (Cmi), the causal agent of bacterial wilt in lucerne, to establish an international reference for the detection of Cmi in lucerne seed. Test plans and kits are submitted to participating laboratories to collect criteria for method validation. Various technologies are considered such as selective media plating, enzym immunoassay (ELISA), metabolic fingerprinting, and polymerase chain reaction (PCR). Test data are reviewed by the ISTA Plant Disease Committee (PDC). Parameters of sensitivity and specificity for the detection of Cmi in lucerne seed are set by the PDC in consultation with regulatory bodies and the seed industry.
