Interpretive Summary:

Technical Abstract: Common bacterial blight [CBB, caused by Xanthomonas campestris pv. phaseoli (Smith) Dye] is a major disease of common bean (Phaseolus vulgaris L.) worldwide. The tepary bean (P. acutifolius A. gray) is known to possess high levels of resistance. The first interspecific cross between common and tepary bean was reported in1956. Eventually the Great Northern Nebraska No.1 (GN#1) cultivar, selected for CBB resistance and good agronomic characteristics, was derived from this interspecific cross in 1961. A later maturing common blight resistant rogue plant found in GN#1 was subsequently named Great Northern Nebraska #1 Sel. 27 (GN sel 27). Evidence that the primary source of CBB resistance in GN sel. 27 might actually derive from Montana No.5 came from two different sources simultaneously. Field trials in North Dakota in 1999 revealed that Montana No.5 exhibited a level of CBB resistance comparable to resistant cultivars with GN sel. 27 derived resistance. At Prosser, the SAP6820 SCAR marker, tightly linked with a maj QTL for CBB resistance derived from GN sel. 27, did not amplify in tepary bean (Table 2) like the other resistance-linked markers. However, SAP6 did amplify in Montana No. 5. To examine the relationship of SAP6 in Montana No.5 with CBB resistance, 50 individual F2 plants from the cross Montana No.5/Othello were assayed for SAP6 marker and inoculated with CBB. Regression analysis revealed that SAP6 did cosegregate (35%) with CBB resistance, confirming that the major-effect QTL in GN sel 27 derives from Montana No.5 not tepary bean.
