Interpretive Summary:

Technical Abstract: A total of 16,424 wheat root expressed sequence tags (ESTs) from the National Science Foundation Wheat EST Genomics Project and the International Triticeae EST Cooperative were annotated. The ESTs were derived from mRNAs expressed in non-stressed root tips, whole roots of seedlings, and roots of tillering plants, or from whole roots and root tips following exposure to drought, salt, and aluminum. BLASTN analysis was used to reduce the redundancy of the collection to 13,721 non-overlapping EST clusters, and BLASTX was used to analyze the functions of the deduced proteins. Under the stringency criteria used, an average of 54% of the deduced open reading frames matched polypeptides with assigned functions, 12% matched hypothetical or expressed proteins with no assigned functions, and 34% showed no matches in the GenBank database. Transposon-like sequences and nodulation-associated transcripts were found among the wheat root ESTs. Abundances and functional diversity of defense-related ESTs expressed in non-stress and stress conditions were compared where possible. Progress in the application of wheat root ESTs for real time-PCR probes and for microarrays will be presented.
