Technical Abstract: The immunogenic effect of outer membrane protein (OMP) of Salmonella enteritidis (SE) in inducing cell-mediated immune response was studied. White leghorn inbred chickens were subcutaneously immunized with OMP or crude protein (CP) from SE, or a commercially SE killed vaccine twice at 6 and 9 wk of age. Proliferation responses of spleen lymphocytes to concanavalin A (Con A), OMP, CP, flagella, porin and lipopolysaccharide (LPS) antigens from SE were evaluated in vitro. Kinetics of serum interleukin (IL)-6 and IL-2 production were evaluated by an indirect enzyme-linked immunosorbent assay (ELISA) at 1, 2, 3, 4, and 5 wk after primary immunization with SE antigens or a SE killed vaccine. OMP- and CP-immunized chickens as well as chickens immunized with a commercial killed vaccine showed a good proliferation response at 4 weeks post primary immunization compared to the control groups which were injected with saline or adjuvant alone. SE flagella antigen was significantly more effective (P < 0.05) in stimulating lymphocyte proliferation in vitro than other antigens. Compared to the control groups, chickens which were immunized with OMP, CP, or killed vaccine produced a significantly higher levels of serum IL-6 and IL-2 (P < 0.05) after primary immunization. These results indicate that a subcutaneous immunization with the killed vaccine, bacterial CP or OMP is capable of eliciting cell-mediated immunity which might be associated with protective immune response to SE.
