Title: DETECTION OF STAPHYLOCOCCAL ENTEROTOXIN B (SEB) WITH SURFACE PLASMON RESONANCE BIOSENSOR

Technical Abstract: An automated and rapid method for detection of staphylococcal enterotoxins is needed by the food industry. We developed a biosensor Sandwich Assay using a surface plasmon resonance (SPR) biosensor for detection of Staphylococcus aureus enterotoxin B (SEB). The assay is based on the immobilization of SEB antibody on a sensor chip followed by a sequential capture the SEB from buffer solutions or samples, and second SEB antibody. The assay conditions were optimized to detect SEB in buffer and ham extracts. The results showed detection of SEB at 2.5 ng/ml in buffer or ham tissue extract. The mean R^2 of 0.982 and 0.995. were determined from the calibration lines of two 8 replicate analyses of SEB spiked in ham extracts. Analysis of 10 and 20 ppb SEB spiked on the surface of ham slices resulted in recoveries of 70 and 87%, and 62 and 86%, respectively. Spiking with 2.5 and 5 ppb SEB had recoveries of 53, 0 and 65, 6%, respectively. The recoveries and detection sensitivity can be improved by modifying the extraction procedure and the use of a specific antibody, respectively. The Biacore analysis is semi-automated and can be developed for multi-toxin detection in foods.
