Interpretive Summary:

Technical Abstract: Basidiomycetes are an important member of fungal soil communities whose population size and diversity are hypothesized to vary with soil type. Though culture-dependent methods may be used to analyze these fungi, variations in nonculturable species would be missed. These organisms are rendered detectable by PCR amplification of Basidiomycete ribosomal DNA. A PCR primer was designed to be specific for Basidiomycetes and useful for community analysis. The developed primer was used to amplify part of the ribosomal small subunit (SSU) and the entire internal transcribed spacer (ITS) region from genomic DNA extracted from pure culture and from soil samples. PCR product from soil was cloned and sequenced, yielding only sequences with nearest neighbors in the Basidiomycetes. Applying Real-time PCR and analysis of terminal restriction fragment length polymorphism (T-RFLP), our primer detected significant differences in Basidiomycete diversity and population size in soils from different land uses.
