Technical Abstract: We are using genomics tools to identify genes involved in disease resistance. As a first step, we utilized results from a large-scale cDNA-AFLP profiling experiment in wheat to determine global gene-expression responses against two fungi, two viruses and Hessian fly. This analysis identified many genes with differential expression specific to each interaction. We used the interaction between wheat and the fungus Mycosphaerella graminicola to further test the hypothesis that many of the host genes showed altered expression in response to infection. Thirty up-regulated genes, such as phenylalanine ammonia lyase, heat-stress genes, transcription factors, peroxidases, etc., were selected for the study. Expression analysis of these genes by quantitative real-time PCR confirmed differential expression in the resistant and susceptible wheat cultivars at several time points after infection. A summary of the findings will be discussed.
