Technical Abstract: Fas-associated death domain protein (FADD) is an adapter molecule expressed in many tissues that mediates cell apoptotic signaling. The full-length sequence of the bovine FADD cDNA was determined by reverse-transcription PCR using mRNA isolated from cultured bovine aortic endothelial cells and cloning of the resulting product into a plasmid vector for DNA sequencing. Sequencing of bovine FADD cDNA revealed approximately 83 and 80% nucleotide sequence identity to its human and murine homologues, respectively. A bovine FADD dominant-negative construct was developed to examine bacterial lipopolysaccharide (LPS)- and TNF-a-induced apoptosis in bovine endothelial cells. GenBank Accession No. AY725483.
