Interpretive Summary:

Technical Abstract: Soybean Cyst nematode (SCN) Heterodera glycines Ichinohe is the most serious pest of soybean worldwide. SCN resistant cultivars are numerous, but nearly all of them trace to a common source of resistance, PI 88788. Soybean plant introduction PI 507354 was recently found to be a unique, yellow seeded resistant source to SCN races 1, 3 and 5 (HG types 2.5.7. 0 and 2-). The objective of this study was to investigate the inheritance of resistance to SCN races 1 and 5 in PI 507354. Hutcheson, susceptible to all known SCN races, was crossed to PI 507354 to generate F1 hybrids, 225 F2 individuals and 225 F2:3 families. Ten seeds in each of the 225 F2:3 families, the two parents, and SCN indicator lines were used in a SCN bioassay to determine resistance. The female index (FI) was calculated to evaluate SCN reproduction on each family. Chi-square (x2) tests were used to test the goodness of fit between observed and expected values for F2:3 data. The data indicated that the response to SCN races 1 and 5 in the F2:3 families was normally distributed; 225 F2:3 progenies showed a good fit to an expected 1 dominant and 3 recessive gene model for resistance to both races 1 and 5. Molecular mapping of QTLs associated with SCN resistance to races 1 and 5 are in progress using SSR and AFLP markers
