Interpretive Summary:

Technical Abstract: Despite increased use and superior performance of PCR assays in clinical laboratories, identification of B. avium still depends upon isolation and biochemical testing. A B. avium-specific PCR has previously been reported, but details critical to reproducing it are not provided and analytical verification was not carried out. We have optimized reaction conditions using the previously described primer set and report 100% assay sensitivity and 98.6% specificity, based on analysis of 72 B. avium isolates and 85 other poultry isolates of various genera and species. The assay can be carried out on purified DNA or on boiled lysates prepared from single colonies. Sequence analysis of the amplicons generated from the B. avium isolates revealed occasional polymorphisms, one of which may be host-specific. The sequence data provides evidence for transmission of B. avium from wildlife to domesticated turkey flocks.
