Interpretive Summary:

Technical Abstract: BAC-derived SSR markers can provide very important information for molecular and physical mapping, and map based cloning and gene sequencing. SSR markers were developed from BAC end sequences of TM-1 (Gossypium hirsutum). The overall objective of this research was to identify chromosomal location of the BAC derived SSR markers in tetraploid cotton. One hundred ninety two BAC-derived SSR primer pairs were screened against five cotton lines [TM-1, DP61, T-701 (Gossypium hirsutum) and, 3-79 (G. barbadense) and G. tomentosum]. Interspecific aneuploid chromosome substitution lines were used to identify chromosomal location of these SSR markers. Overall, 56.25% of these BAC-derived SSR primers were polymorphic at the interspecific level. 39.58% are polymorphic between TM-1 and 3-79, 30.72% are polymorphic between TM-1 and G. tomentosum. 21.35% are polymorphic between TM-1, 3-79 and G. tomentosum . Our preliminary results revealed the chromosomal location of several of these BAC derived SSR markers. The detailed results will be presented at the meeting.
