Interpretive Summary:

Technical Abstract: Soybean Cyst Nematode (SCN) is the most important soybean pest throughout the world. Soybean plant introduction (PI) 494182 is a new source of resistance to SCN races 3 and 5 from Japan. The objective of this study was to find quantitative trait loci (QTLs) for resistance to SCN race 3 in PI 494182. The mapping population consisted of 162 F3:4 lines from a cross between Skylla x PI 494182. The population was genotyped with 107 polymorphic SSR markers from 17 Linkage Groups (LG) and was tested for resistance to SCN races 3. Both single marker analysis (SMA) and composite interval mapping (CIM) methods were used to locate the QTLs. With SMA, 11 markers from LGs A2, B2, F, G, and K were found significantly in association with resistance to SCN race 3. In the CIM analysis, a significant QTL was found on LG G near the rhg1 gene.
