Interpretive Summary: A series of laboratory manuals are in the process of being written for serious students of technological exploitation of important microorganisms. The series is divided into two parts, bacteria and fungi. Scientists at RRC were invited to write the chapter dealing with the isolation, growth measurement and transformation of a species of bacteria that offers biocontrol of a pathogenic fungus of corn, Fusarium verticillioides. The work is based on the exciting world of bacterial endophytes using the pioneering work dealing with the initial isolation and characterization of the patented bacterium Bacillus mojavensis. In addition to all aspects of manipulating this bacterium in the laboratory, valuable techniques such as characterizing specific strains based on nutritional and antifungal qualities are described as well techniques describing how to genetically alter this bacterium with specific fluorescent genes useful for measuring and following it during ecological and field studies on its antagonism to fungi.
