Interpretive Summary:

Technical Abstract: Marek’s Disease is a T cell lymphoma disease of chicken induced by an oncogenic, cell associated alpha herpes virus. Oncogenicity in the Marek’s disease is mostly attributed to a transcription factor termed as Meq. To understand the mechanisms of oncogenicity of Meq, it is necessary to understand its interactions with the chicken genome. To study how Meq affects gene expression in the host genome we are examining both protein-DNA and protein-protein interactions. The focus of the present study was on the interaction of Meq with other transcription factors. We used a MDV-transformed T cell line (MSB-1) in which MDV is latent. In our first experiment, we hypothesized that Meq expression changes with time upon reactivation by bromodeoxyuridine. The levels of Meq were quantitated at 0, 24, 48, 96 hrs after reactivation. Next, protein fractions bound by Meq were enriched by chromatin immunoprecipitation using anti-Meq antibody. By using in-solution tryptic digestion followed by mass spectrometric analysis, a number of unique proteins were discovered. The direct binding interactions between Meq and the target proteins will then be evaluated using PyMOL analysis.
