Interpretive Summary:

Technical Abstract: The current study was conducted to investigate the effects of the direct-fed microbials (DFM) including three Bacillus subtilis strains on the modulation of transcriptional profile in chicken intestinal intraepithelial lymphocytes (IEL). The multiple-strain DFM product modified 453 probes from 1,985 microarray elements with above 2.0-fold up- or down changes in chicken IEL when they’re compared with non-treated controls and those modulations were closely associated with the biological functions of lipid metabolism and the interaction between pro-inflammatory cytokine activated by lipopolysaccharide (LPS) and lipid metabolic modulation. Macrophage activation induced by the DFM treatment was also accessed by measuring nitric oxide (NO) levels in cultures of LPS-stimulated splenocytes. The results suggested that the uptake of multi-strain DFM augment the innate immune system against the infection by enteric pathogens. Future studies to identify novel functions and/or potential anti-pathogenic effects of DFMs as the feed additives based upon these results may now be feasible.
