Title: Aerobic growth of campylobacter in media supplemented with a-ketoglutaric, lactic, and/or fumaric acids

Interpretive Summary: A series of experiments were performed to determine if cultures of Campylobacter bacteria can be grown under aerobic (atmospheres composed of normal breathing air) conditions. Presently, microbiologists grow these bacteria in closed jars under microaerobic (atmospheres with reduced oxygen concentrations) conditions. Recent experiments have indicated that growth of Campylobacter is stimulated in media supplemented with selected organic acids; therefore, these experiments were conducted to determine if Campylobacter that are provided some of these acids can also grow under aerobic conditions. Bacterial growth media was supplemented with combinations of three organic acids, a-ketoglutaric, lactic, and/or fumaric acids. Supplemented media was inoculated with Campylobacter coli, Campylobacter fetus, or Campylobacter jejuni, and then incubated aerobically at 37 degrees Celsius for 72 hours. After incubation, increased turbidity of culture media was measured to determine bacterial growth. Results indicated that there was no growth of either Campylobacter in media with no organic acids or media supplemented with only a-ketoglutaric acid. On the other hand, there was growth of C. fetus in media supplemented with only lactic acid. Furthermore, there was additional growth of C. fetus, as well as growth of C. coli, in media supplemented with only fumaric acid. The greatest growth of all 3 Campylobacter was produced in media containing a mixture of fumaric and lactic acids or a mixture of a-ketoglutaric, lactic, and fumaric acids. This study indicates that Campylobacter can be grown under aerobic conditions in media supplemented with selected organic acids.

Technical Abstract: A study was conducted to examine the ability of Campylobacter spp. to grow aerobically in media supplemented with selected organic acids. Basal broth media composed of tryptose, yeast extract, and a mineral-vitamin solution was supplemented with a-ketoglutaric, lactic, and/or fumaric acids. The final pH of the media was 6.5. Media was inoculated with approximately 106 colony-forming-units/ml of Campylobacter coli ATCC 33559, Campylobacter fetus ATCC 27349, or Campylobacter jejuni ATCC 33560. Inoculated media was incubated aerobically at 37C for 72 h in a Bioscreen C Microbiology Reader. During incubation, the optical density (OD) of cultures was measured at 600 nm. After incubation, statistical analysis of final culture OD was performed to determine significant differences in culture growth. Results indicated that there was no significant (P < 0.05) growth of either isolate in non-supplemented media or media supplemented with only a-ketoglutaric acid. In contrast, there was significant growth of C. fetus in media supplemented with only lactic acid. Furthermore, there was additional growth of C. fetus, as well as growth of C. coli, in media supplemented with only fumaric acid. However, greatest growth of each isolate was produced in media supplemented with a mixture of fumaric and lactic acids and a mixture of a-ketoglutaric, lactic, and fumaric acids. Findings of this study indicate that Campylobacter spp. can grow aerobically in media supplemented with selected organic acids.
