Technical Abstract: Fumonisins, toxic metabolites of certain Fusarium molds, can be found in corn and corn-based foods at levels sufficient to cause disease in horses and swine. A variety of immunochemical methods for the intact fumonisins have been developed based upon polyclonal and monoclonal antibodies. Affinity columns and sensitive ELISAs are now available commercially for the intact fumonisins. Anti- idiotype and anti-anti-idiotype antibodies have also been produced. Recently we reported the development of two sets of monoclonal antibodies with differing specificities; the first (P2A5-3-F3) bound intact fumonisins while the second (P2F11-3-H7) bound their hydrolysis products. These antibodies have been used for ELISAs and have been combined into affinity columns for the cleanup of corn products where the presence of both sets of compounds is suspected. The antibodies have also served as the basis for the development of a fiber-optic immunosensor for the fumonisins, in which unlabeled fumonisin B1 (FB1) competes with fluorescein isothiocyanate labeled FB1 for binding to P2A5-3-F3 antibody covalently coupled to a fiber-optic probe.
