Technical Abstract: The maize virus program at Wooster, OH USA has been a cooperative research effort between Federal and State scientists to control maize viruses since the mid-1960's. In the early years, virus identification and characterization, breeding for disease resistance/tolerance, and maize virus epidemiology were emphasized. Recently, the emphasis has been to develop an understanding of the mechanisms of virus-host interactions at the molecular level and from this understanding, devise innovative methods for disease control. In this presentation, the traditional strategies for disease control augmented by the tactics of molecular biology to elucidate the genetic basis for resistance to maize virus diseases will be reviewed. The impact of variability in virus and host populations on the evaluation of disease resistance/tolerance; methodologies used to identify virus-host variations and to screen for disease resistance/tolerance; and the application of techniques from molecular biology to identify virus disease resistant genes in maize also will be discussed.
