Interpretive Summary:

Technical Abstract: Molecular markers associated with tolerance in oats to barley yellow dwarf virus (BYDV) infection were identified using four pairs of near isogenic lines (NILs) differing in their responses to BYDV infection. Total oat DNA was extracted from each of the NILs and the parents used to derive the NILs and subjected to amplified fragment length polymorphism analysis. The DNAs were restricted with EcoRI and MseI, ligated to adapters and selectively amplified using 64 different combinations of primers. DNA fragments were separated on high resolution denaturing DNA sequencing gels and detected by autoradiography. Each primer combinations produced an average of 94 bands, approximately 10% of which were polymorphic between the parents. Only two primer combinations produced banding patterns that were associated with tolerant but not sensitive NILs.
