Technical Abstract: There is no comprehensive laboratory guidebook to provide the instructions for AI technology and fertility evaluation techniques commonly used by the poultry industry and poultry scientist in academia. Therefore, the objective of this text is to provide a comprehensive, user friendly, laboratory manual which provides a step-by-step approach to the more commonly used semen evaluation and storage procedures. In addition, procedures for some of the more novel techniques developed to assess semen quality and fertility are also described in detail. Before attempting a laboratory procedure, we recommend the following: study the directions carefully; plan your work in advance, checking to see that all reagents and materials are available; keep notes of what you have done as calculation errors can be easily corrected if your notes and data are adequately recorded; and finally, the data sheets included with some of the laboratory yprocedures are meant as a guide, and should be modified to suit your laboratory's needs.
