Title: TRANSCRIPTIONAL REGULATION OF IL-1 RECEPTORS IN BOVINE LEUKOCYTES IN VITRO AND IN VIVO

Technical Abstract: There is very limited information regarding the regulation of interleukin-1 receptor (IL-1R) gene expression in leukocytes of food animal species. In the present study, we investigated the in vitro regulation of bovine type I (IL-1RI) and type II (IL-1RII) gene expression in bovine leukocytes in response to the cognate ligand rBoIL-1beta, other bovine cytokines (rBoGM-CSF, rBoIL-4, and rBoIFN-gamma), and dexamethasone. Our findings suggest that anti-inflammatory agents, such as dexamethasone and L-4 increased the expression of IL-1RII in bovine peripheral blood mononuclear cells (PBMCs) and neutrophils (PMNs) as determined by Northern analysis. In contrast, the TH1 cytokine rBoIFN-gamma down-regulated the expression of both IL-1RI and IL-1RII in bovine PBMCs and PMNs as assessed by modified semi-quantitative RT-PCR. rBoIL-1beta up-regulated IL-1RII mRNA in bovine PBMCs and PMNs, whereas, in contrast, IL-1RI was up-regulated in PBMCs and down-regulated in bovine PMNs by rBoIL-1beta. The hematopoietic growth factor, rBoGM-CSF, up-regulated both IL-1RI and IL-1RII mRNAs in bovine PBMCs and PMNs, as determined by Northern analysis. In vivo studies are being performed to confirm the effects of dexamethasone on IL-1R expression noted in vitro.
