Interpretive Summary: The authors have demonstrated the isolation of a cDNA for an important regulatory protein from a lower plant model system. The clone, for CDPK, has motifs and structure similar to higher plant homologs. This demonstrates the usefulness of using bryophytes as model systems and the utility of the genetic components that can be isolated from them. This particular clone has important possible utility in the study of plant stress responses since the gene for which the cDNA represents appears to be a rehydrin (genes involved in the response of mosses to extreme water stress).

Technical Abstract: This manuscript describes the isolation, for the first time, of a bryophyte clone for an important signaling protein Calmodulin-like Domain Protein Kinase (CDPK). The clone was isolated from a moss library using a PCR generated fragment of a CDPK isolated from Arabidopsis thaliana. The full length clone is 2243bp and encompasses an open reading frame that is 1737 base pairs long. This open reading frame encodes a predicted CDPK of 578 amino acids. The protein contains the four domain structures characteristic of CDPKs. The moss CDPK has a very long (111 amino acid) N-terminal variable domain. The adjacent kinase catalytic domain contains the conserved structural motifs common to all members of the eukaryotic kinase superfamily.
