Technical Abstract: Pasteurella haemolytica (PH) colonization of the upper respiratory tract was inhibited in experimentally-vaccinated cattle. To determine possible relationships between antibody and protection, serum and nasal secretion pools from the cattle were analyzed by Western blots against an outer membrane protein (OMP) preparation containing iron-regulated proteins. The eOMP was prepared from PH grown in BHI broth with 150 uM 2,2-dipyridyl and supplemented with 20 ug of bovine holo-transferrin/ml. Iron-regulated OMP were represented by bands of approximately 80 and 75 kDa. Vaccinate serums had strong reactions with both iron-regulated OMP, while nonvaccinate serums had very weak reactions to them. Western blots of both vaccinated and nonvaccinated nasal secretion pools showed weak reactions with proteins of approximately 41 and 39 kDa. Serums from one vaccinated but colonized calf, and from one unvaccinated but uncolonized calf reacted the same as serums from the other vaccinates and nonvaccinates respectively. Vaccination with the experimental vaccine elicited serum antibody to iron- regulated OMPs. The antibodies may reflect the ability to inhibit colonization of the upper respiratory tract.
