Technical Abstract: Recent potato late blight epidemics have resulted in substantial yield losses in some areas of the US. To expedite the development of cultivars with resistance to newly emerging genotypes of the causal fungus, Phytophthora infestans, we have initiated a project to tag LBR genes with molecular markers. An F1 segregating population with 276 individuals was produced from a cross between resistant and susceptible lines selected from a diploid Solanum phureja x S. stenotomum hybrid population based on field trials and the detached leaf assay. The parental lines were screened for DNA polymorphism using published RFLP markers selected to ensure good coverage of the genome and the resistance gene analogs isolated from the resistant parental line by PCR cloning. Of 44 DNA probes surveyed thus far, 93% (41) detected polymorphism between the parental lines with at least one of the five restriction enzymes DraI, EcoRI, HindIII, XbaI and TaqI. The high level of DNA polymorphism would greatly facilitate molecular tagging of LBR genes in the F1 segregating population.
