Interpretive Summary:

Technical Abstract: The Nartia strain of citrus tristeza virus (CTV) has been widely used during last fifty years for cross protection against stem pitting CTV in South African grapefruit. We have previously shown the presence of several genotypes of CTV in this isolate by use of a heteroduplex mobility assay of a 400 bp region (nt 1074-1478) using primers which amplify a product in all CTV isolates tested. Single aphid transmitted (SAT) isolates of Narita show a wide array of symptoms as well as serological diversity. CTV populations of the source and the SAT plants were analyzed by RT-PCR to amplify the 400 bp region, then cloned and the clones screened by PCR using CTV genotype-specific primers. Multiple populations of CTV were present in most of the SAT isolates, but in different ratios which may be responsible for the phenotypes differences and serological diversity.
