Interpretive Summary:

Technical Abstract: The majority of PCR-based markers mapped in carrot (Daucus carota) have been either random amplified polymorphic DNA (RAPD) markers or amplified fragment length polymorphism (AFLP) markers. RAPD markers suffer from poor reproducibility and both RAPD and AFLP primarily produce dominant markers. A recent method, sequence related amplified polymorphism (SRAP), has been reported to produce up to 20% co-dominant markers with distribution and reproducibility similar to AFLP. We developed SRAP markers linked to the Mj-1 locus, which confers resistance to the root-knot nematode Meloidogyne javanica, in carrot. The SRAP markers were mapped in a population previously used for mapping AFLP and RAPD markers linked to the Mj-1 locus. The utility of using SRAP markers in closely related carrot cultivars is discussed and their ease of use and reproducibility are compared with AFLP.
