Interpretive Summary: Technical Abstract: Currently, levels of obesity in Middle Eastern countries are increasing. Phytochemicals have anti-obesogenic properties as evidenced by prevention of adipocyte differentiation. In Yemen, Pulicaria jaubertii E.Gamal-Eldin (PJ) is a food additive and a traditional medicine. We tested the ability of extracts derived from PJ to inhibit the differentiation of 3T3-L1 preadipocytes. Methanolic extracts of PJ blocked triglyceride accumulation and leptin expression, but not expression of fatty acid binding protein 4 (FABP4), indicating that methanolic extracts of PJ block triglyceride accumulation but not adipocyte differentiation. Previous data demonstrate that elevation of NADPH:quinone oxidoreductase 1 (NQO1) blocks triglyceride accumulation. Treatment of 3T3-L1 preadipocytes increased NQO1 content in part as a response to glutathione (GSH) depletion. PJ treatment increased the expression of glutamate cysteine ligase catalytic subunit and glutathione reductase suggesting that PJ activates the anti-oxidant response element pathway. Further fractionation of the methanolic PJ extract with hexane and dichloromethane (DCM) showed that the DCM fraction, but not the hexane fraction, recapitulated the effects of the methanolic extract. GC-MS analysis of the fractions revealed multiple compounds including carvotanacetone as most abundant in hexane fraction. Occurrence of catechin-like polyphenolics was proposed based on GC-MS and LC with high resolution MS analysis. Funding: USDA-ARS and the Fulbright Visiting Scholar Program. |
