Interpretive Summary:

Technical Abstract: Pasteurella haemolytica biotype A serotype 1, a Gram-negative organism, is the major cause of bovine pneumonic pasteurellosis resulting in major economic loss to the cattle industry. It has been reported that cattle that have recovered from natural infection appear to be resistant to subsequent infection, implying that an effective vaccine could be developed dif the immunogens responsible for the natural protection could be identified. A method for the preparative scale isolation of these immunogenic proteins is described. An immunoaffinity column was prepared from the IgG fraction of serum from calves actively infected with P. haemolytica. An outer membrane protein-rich fraction was obtained from sonicated salt-extracted P. haemolytica cells by extraction with N-lauroyl sarcosinate (Sarkosyl) and the Sarkosyl-insoluble proteins were subjected to immunoaffinity chromatography. Three proteins with molecular weights of f55, 92, and 105 kDa were isolated as determined by SDS-PAGE under denaturing conditions. These three proteins represent the immunogenic outer membrane immunogens of P. haemolytica associated with pasteurellosis of cattle.
