Interpretive Summary: Technical Abstract: ApoE knockout (ApoE-/-) mice fed AIN-93G diet (CD) formulated to contain 1 % freeze-dried whole wild blueberries (CD1 percent BB) were found to have significantly less atherosclerotic lesions in aorta. Biomarkers of lipid peroxidation, including F2-isoprostanes, hydroxyoctadecadienoic acids (HODEs) and hydroxyeicosatetraenoic acids (HETEs) were measured in serum. 8-HETE, 12-HETE, and 13-HODE were found to be significantly lower in serum of CD1 percent BB fed animals. Gene expressions of 17 major antioxidant/oxidant enzymes in aorta were further measured by PCR array. In pooled samples, three antioxidant enzymes, superoxide dismutase 2, glutathione reductase, thioredoxin reductase 1, were found to be up-regulated, and one antioxidant enzyme catalase was found to be down-regulated in CD1percentBB fed animals. These results were further verified by Real-Time PCR in individual samples. Antioxidant activities of BB, especially the ability to inhibit lipid peroxidation and increase certain antioxidant enzymes may partly contribute to the protective effects of BB against atherosclerosis. |
