Objective:
The objectives of this proposal are to enhance Botrytis, and related species, management through enhanced detection and monitoring of fungicide resistance development. 1) Develop molecular markers for fungicide resistance to FRACs 2, 7, 9, 11, and 12. 2) Validate and deploy fungicide resistance markers for California specialty crops.

Approach:
For objective 1, a transcriptomics-based time course experiment will be used for identifying targets associated with fludioxonil resistance. DNA-based markers will be developed using identified genes. An antibody-based assay will be used for FRAC11 resistance detection. The antibody has already been developed, and various buffers will be tested for ability to lyse spores and allow antibody to bind. Cross resistance will be evaluated for FRAC 7 to ensure developed markers can effectively detect resistance from different FRAC 7 products. For objective 2, tools developed in objective 1 will be deployed in field and tested for use in other crops and with other fungal species to test for potential cross-reactivity and transferability to other common pathogens including Cercospora and Alternaria.
