Research Project: Transcriptome and Proteome Analysis of Barley in Response to Abiotic Stress and During Malting

Location: Cereal Crops Research

Project Number: 5090-21430-012-003-S
Project Type: Non-Assistance Cooperative Agreement

Start Date: Apr 1, 2022
End Date: Sep 30, 2023

Objective:
A large scale RNA-seq data set has been collected from two barley lines, one (stress sensitive) and the other (resistant) which were exposed to heat, drought or combined heat and drought stress. Co-operator will conduct the RNA-seq analysis of this large data set using bioinformatic tools.

Approach:
Map the sequenced reads from 140 different barley samples to the latest version of the barley Morex version 3 reference genome. Conduct in-depth quality analysis of the libraries, make comparisons to identify differentially expressed genes in the two barley lines and also undertake thorough gene ontology and pathway analysis. Retrieve the latest gene identifiers from Morex Version 3 genome assembly and their associated ontologies for the transcriptome and proteome data sets associated with different stages of malting barley previously published by ARS scientists.
