Research Project: Evaluate and Compare Current Amylose Quantification Methods for Grain Sorghum

Location: Grain Quality and Structure Research

Project Number: 3020-43440-002-028-S
Project Type: Non-Assistance Cooperative Agreement

Start Date: Sep 1, 2022
End Date: Aug 31, 2025

Objective:
Develop a quick and reliable method to measure amylose contents in sorghum grain and understand how the results from the method are compared with those from other methods.

Approach:
1. Identify and collect sorghum varieties and breeding lines for the project. 2. Develop a quick and easy screen method to detect contamination of normal sorghum grains in waxy grain. 3. Evaluate and compare different methods (potentiometric, colorimetric, concanavalin A, GPC/SEC, and DSC) for determining amylose contents in sorghum grain/flour. 4. Evaluate GxE effects on amylose contents in grain sorghum and their functionality in food applications.
