Objective:
Objective 1: To sequence the transcriptome of lettuce roots in plants under thrip and Impatiens necrotic spot virus (INSV) pressure in order to develop testable hypotheses for the molecular mechanisms involved in biotic stress response.
Approach:
We will use inoculation methods developed by collaborators using viruliferous thrips under controlled conditions. We will sample roots from a minimum of 3 plants of each of 4 cultivars with varying susceptibility to thrips and INSV under 3 treatments (controls that are untreated with thrips and INSV, plants that are treated with thrips only, and plants that are treated with viruliferous thrips), at 4 time periods after inoculation (12 hours post treatment, 24 hours post treatment, 72 hours post treatment, and 96 hours). Tissue will be taken from roots, and submitted to transcriptome sequencing. Sampling is destructive, so new plants will be used for each time period sample.
