Research Project: Developing Stem Rust Resistant Barley Germplasm

Objective:
The overall goal of this project is to develop agronomically advanced malting barley cultivars with broad-based resistance to stem rust.

Approach:
The objective will be accomplished by: 1) developing mapping populations derived from new sources of adult plant resistance; 2) elucidating the number, chromosomal position, and effects of loci conferring adult plant resistance in these sources; 3) developing molecular markers for previously described genes conferring all-stage (Mendelian) and also for those conferring adult plant resistance from newly identified sources; 4) assessing the implementation of marker screening on early generation (F3) breeding material to select stem rust resistant lines for advancement to yield trials; and 5) selecting and releasing new barley germplasm with enhanced stem rust resistance. Four mapping populations will be developed from accessions exhibiting the highest level of adult plant resistance from evaluations conducted in Africa in 2015. Each of these resistance donors will be crossed with a susceptible barley line (PI 60693) to ultimately generate recombinant inbred lines (RILs) via single-seed descent. RILs will be selfed to at least the F6 generation before genotyping and phenotyping.
