Research Project: Locating Pre-Harvest Sprouting Resistance in Elite Wheat Breeding Lines

Objective:
1) Survey pre-harvest sprouting (PHS) resistance on a portion of breeding lines from multiple state wheat breeding programs under development as new varieties chosen for greatest genetic variation. 2) Genetic analyses performed to find genes or chromosome locations involved in PHS resistance. 3) Chromosome locations assessed for potential as new markers of PHS resistance.

Approach:
Objective 1: ~800-1000 lines will be chosen from all breeding lines genotyped in seven state breeding programs based on genetic diversity measured by SNP markers covering the entire genome provided by the collaborator. PHS will be measured for all three years. PHS will be measured using AACCI methods (AACC International 2010) for alpha amylase activity (AACCI method 22-02.01) modified for high throughput using a method developed in by ARS and falling number (AACCI method 56- 81.03). Objective 2: Genome-Wide Association Studies (GWAS) will be performed on the lines for all three years using markers provided by the collaborator. Depending on marker density, a subset of markers may be used for the initial screening to attain the best power to detect. Positive correlations will be reduced in size using the full set of markers in the selected area. Objective 3: Locations or genes found to be significant for PHS resistance in Objective 2 or published for wheat or barley will be used to develop markers. These markers will be tested on either the lines in this study (if they are published markers) or a previously developed historic diversity panel for which PHS data of three plus years is available to determine reliability of the markers. The marker analyses will be made available to all wheat breeders.
