Objective:
Characterize the substrate specificity of the Fg effector proteases. Identification of cleavage site sequences for each of the candidate proteases will be determined using a high-throughput, yeast-based system. In parallel, we will also test whether the candidate Fg proteases are required for virulence.
Approach:
The cooperator will utilize bioinformatics to identify candidate effector proteases secreted by Fusarium graminearum during pathogenesis. Knockout mutations with then be created in the candidate fungal effector protease genes and the resultant mutants will be tested for loss of virulence in Fusarium head blight assays.
