Objective:
1. Develop and characterize panels of monoclonal antibodies (mAb) specific for swine immune proteins, e.g., cytokines.
2. Generate swine leukocyte antigenclass I tetramers for several common US pig breeds
3. Express specific transcription factors regulating critical immune interactions and develop/identify mAbs to them
4. Generate/identify mAbs specific for porcine cluster differentiation (CD) antigens
5. Identify commercial partners for downstream use of developed reagents.
Approach:
ARS will focus on Objectives 3 and 4.
Objective 3 and 4 targets will include characterizing mAbs produced by commercial partners. This may also include analyses of mAbs produced during a previous collaboration. Western blots and flow cytometry will be used to confirm specificity of mAbs, define epitopes recognized and cross species reactivities. Intracellular staining will be tested on pig cell subsets as appropriate.
