Objective:
Utilize metabolome and phytohormone analyses to have a better understanding of molecular mechanisms associated with postharvest wound-healing, tuber dormancy and sprout growth processes.

Approach:
Metabolome and phytohormone profiling will be performed to investigate the underlying molecular mechanisms that regulate tuber dormancy and wound healing processes of potato. Experiments will be conducted in a multi-institutional research setting, and the cooperator will carry out the following: 1. Provide expertise on biochemical assays, and optimize the protocols for potato tissues. 2. Ensure the outlined biochemical assays are completed in a timely manner for the outlined potato physiology experiments. 3. Process the tissue samples to obtain metabolome profiling (monoterpenoid volatiles, polar amino acids, or hydrophobic lipids), and phytohormone profiling (auxin, abscisic acid, cytokinins, gibberellic acid, salicylic acid, and jasmonic acid). 4. Analyze the metabolome and phytohormone profiling data, and help integrate the results with relevant transcriptome data to explain the molecular mechanisms controlling potato tuber dormancy and wound healing. 5. Contribute preparation of a final report to distribute the results.
