Research Project: Stacking RNAi-based Resistance to Phytophthora, Crown Gall, and Nematodes in Almond and Walnut Rootstocks

Location: Crops Pathology and Genetics Research

Project Number: 2032-22000-017-050-A
Project Type: Cooperative Agreement

Start Date: Nov 1, 2022
End Date: Feb 28, 2025

Objective:
Develop walnut and almond RNAi lines for resistance to the three main soilborne diseases (Phytophthora root and crown rot, crown gall, and root-lesion nematodes)

Approach:
Cooperator will construct transformation vectors for RNAi against Phytophthora, then co-transform somatic embryos with two vectors, which confer resistance against Phytophthora and crown gall (the vector already available). Cooperator will subsequently propagate transgenic embryo lines, inoculate them with pathogens and evaluate resistance. Transgenic lines showing resistance will then be propagated, rooted, and grown in pots. Cooperator will conduct greenhouse trials for Phytophthora and crown gall resistance.
