Objective:
Phenotype near-isogenic introgression peanut lines for seed colonization by Aspergillus flavus.

Approach:
Seed colonization assays will be conducted in the lab, using a non-toxigenic isolates of Aspergillus flavus on seed with and without testa. Colonization will be quantified using ImageJ or similar software. Transcript abundance of the regulatory genes for aflatoxin production will also be measure using RT-PCR. Differences among selected near-isogenic introgression lines for early leaf spot resistance will be screened through RNA sequencing.
