Research Project: Gene Expression Changes in Primary Swine Cells in Response to L929 Conditioned Media

Objective:
Replication of African Swine Fever Virus in primary cells is enhanced by treatment of cells with media containing L929 conditioned media as a component. To better understand this process we seek to work with the cooperator to perform deep sequencing to evaluate changes in gene expression in swine primary cells following treatment with L929 conditioned media.

Approach:
ARS scientists will collect swine blood and perform extraction of primary cells capable of sustaining African Swine Fever Replication such as buffy coat cells or peripheral blood mononuclear cells and perform treatment of the cells with L929 conditioned media. Following a 24 hour incubation in L929 conditioned media ARS scientists will perform RNA extraction and ship purified RNA to the cooperator. Cooperator will perform sequencing on RNA samples to identify transcripts and perform comparison with control samples, also provided by ARS, to identify changes in gene expression induced following incubation in L929 conditioned media.
