Research Project: Advancement of Disease Resistant Germplasm using Quantitative Genetics and Marker Assisted Selection

Objective:
There are two main objectives for this project. The first is to identify parental lines and parental line combinations that provide superior yield (and quality) on average relative to other potato clones crossed with germplasm containing Columbia root-knot nematode resistance. This will be evaluated by estimating the general and specific combining ability of individual clones and clone combinations. The second objective is to identify clones containing both Potato virus Y and Tobacco rattle virus resistance from segregating populations using marker assisted selection.

Approach:
To achieve the first objective, 30 breeding families will be planted and harvested (10 russet clones crossed to 3 Columbia root-knot nematode resistant clones) and ]their performance will be evaluated using the potato grading capabilities managed by Washington State University at the potato research facility in Prosser, Washington. To achieve the second objective 1000 seedlings from a Castle Russet X A06084-1TE family will be grown out and genotyped for Potato virus Y and Tobacco rattle virus resistance using KASP markers. Individuals that contain both PVY and TRV resistance alleles will evaluated for further selection, whereas those containing one resistance marker and those carrying no resistance marker will be used to evaluate the efficiency of selection through trial in tobacco rattle virus and/or potato virus Y disease fields.
