Objective:
Investigate the pathogenesis of novel swine coronaviruses focusing on viral determinants affecting replication and host immunomodulation.

Approach:
An infectious swine acute diarrhea syndrome coronavirus (SADS-CoV) clone will be constructed. The pathogenicity of the virus derived from this clone will be evaluated in swine at ARS. Attenuating mutations previously demonstrated in another swine enteric coronavirus (porcine epidemic diarrhea virus) will be applied to the SADS-CoV. Attenuation of the SADS-CoV and potential for vaccine use will be confirmed through in vivo study.
