Objective:
1. Observe and measure the permeation of cryoprotectant solutions into plant cells and shoot tips. 2. Identify the concentrations, permeation rates, and subcellular localization of cryoprotectants in plant cells.

Approach:
Research will be performed using state-of-the-art vibrational microscopy to determine cryoprotectant permeability in plant cells and shoot tip systems. Raman spectroscopy, light microscopy, and confocal microscopy will be used. These data will be using in conjunction with USDA transmission electron micrographs to determine subcellular compartmentalization of cryoprotectants within cells. Ultimately, the techniques will show the location and translocation of plant cryoprotectants that are critical for successful long-term cryopreservation of clonally propagated materials in the National plant Germplasm System.
