Research Project: Genomic Analysis of Pest Resistance Genes in US Winter Wheat

Location: Hard Winter Wheat Genetics Research

Project Number: 3020-21000-012-020-S
Project Type: Non-Assistance Cooperative Agreement

Start Date: Sep 1, 2023
End Date: Aug 31, 2027

Objective:
1. Identify and validation of wheat chromosome regions harboring QTLs for rust and Hessian fly resistance 2. Develop selectable markers for rust and Hessian fly resistance 3. Analyze the whole genome sequence data of US winter wheat to characterize wheat resistance to various pests

Approach:
1. Advance three populations derived from the wheat accessions that carry three different Hessian fly (HF) resistance genes. 2. Phenotype the F4 populations in greenhouse for HF resistance. 3. Genotype the populations using GBS markers to determine the regions harboring resistance genes/QTLs. 4.Finely map the genes/QTLs by adding more markers derived from exome capture and selecting new recombinants from secondary populations developed from NILs. 5. Sequence, assembly and annotate the whole genomes of two winter wheat cultivars with target traits using PacBio HiFi sequencing technology and identify the candidate genes in the target chromosome regions.
