Objective:
1. Establish a Nextstrain-based, open-access system integrating viral genomic sequences with epidemiological data for real-time tracking and surveillance of Grapevine leafroll-associated virus 3 (GLRaV-3), the main etiological agent of grapevine leafroll disease (GLD), to advance disease management strategies in the Pacific Northwest.
Approach:
Complete and near-complete genome sequences, as well as the coat protein (CP) sequences, of GLRaV-3 generated from the on-going NCSFR-funded project (Tracking
Genetic Variants of Grapevine Leafroll-Associated Virus 3 for Disease Prevention and Control in Pacific Northwest Vineyards) and retrieved from the GenBank database will be used to prepare multiple sequence alignments using established protocols in Washington State University's lab. All sequence data will be organized in a metadata table, with information on virus isolate, grapevine cultivar, region of the virus collection, and other pertinent information. The GLRaV-3 Nextstrain builds for near-complete genomes and CP sequences of GLRaV-3 will be deposited on the GitHub domain maintained by USDA-ARS.
